L\u00e3o h\u00f3a n\u1ed9i sinh (tu\u1ed5i sinh h\u1ecdc) \u0111\u01b0\u1ee3c th\u00fac \u0111\u1ea9y b\u1edfi qu\u00e1 tr\u00ecnh di truy\u1ec1n v\u00e0 chuy\u1ec3n h\u00f3a, trong khi l\u00e3o h\u00f3a ngo\u1ea1i sinh di\u1ec5n ra nhanh h\u01a1n do \u1ea3nh h\u01b0\u1edfng c\u1ee7a m\u00f4i tr\u01b0\u1eddng, bao g\u1ed3m b\u1ee9c x\u1ea1 tia c\u1ef1c t\u00edm (UV), \u00f4 nhi\u1ec5m, h\u00fat thu\u1ed1c v\u00e0 m\u1ea5t c\u00e2n b\u1eb1ng dinh d\u01b0\u1ee1ng. Trong s\u1ed1 \u0111\u00f3, vi\u1ec7c ti\u1ebfp x\u00fac k\u00e9o d\u00e0i v\u1edbi \u00e1nh s\u00e1ng UV\u2014\u0111\u1eb7c bi\u1ec7t l\u00e0 UVA (315\u2013400 nm) v\u00e0 UVB (290\u2013315 nm)\u2014\u0111\u01b0\u1ee3c c\u00f4ng nh\u1eadn r\u1ed9ng r\u00e3i l\u00e0 nguy\u00ean nh\u00e2n ch\u00ednh g\u00e2y l\u00e3o h\u00f3a da (photoaging). L\u00e3o h\u00f3a da do \u00e1nh s\u00e1ng \u0111\u1eb7c tr\u01b0ng b\u1edfi c\u00e1c n\u1ebfp nh\u0103n s\u00e2u, kh\u00f4 da, thay \u0111\u1ed5i c\u1ea5u tr\u00fac b\u1ec1 m\u1eb7t v\u00e0 r\u1ed1i lo\u1ea1n s\u1eafc t\u1ed1, d\u1eabn \u0111\u1ebfn nh\u1eefng quan ng\u1ea1i h\u00e0ng \u0111\u1ea7u c\u1ea3 v\u1ec1 m\u1eb7t th\u1ea9m m\u1ef9 l\u1eabn y khoa. C\u00e1c li\u1ec7u ph\u00e1p truy\u1ec1n th\u1ed1ng, bao g\u1ed3m retinoid b\u00f4i ngo\u00e0i da v\u00e0 nhi\u1ec1u th\u1ee7 thu\u1eadt th\u1ea9m m\u1ef9, \u0111\u00e3 \u0111\u01b0\u1ee3c s\u1eed d\u1ee5ng nh\u1eb1m gi\u1ea3i quy\u1ebft nh\u1eefng v\u1ea5n \u0111\u1ec1 n\u00e0y nh\u01b0ng th\u01b0\u1eddng g\u00e2y k\u00edch \u1ee9ng da, m\u1ea9n \u0111\u1ecf ho\u1eb7c bong tr\u00f3c. Do \u0111\u00f3, l\u0129nh v\u1ef1c da li\u1ec5u v\u1eabn ti\u1ebfp t\u1ee5c t\u00ecm ki\u1ebfm c\u00e1c ph\u01b0\u01a1ng ph\u00e1p m\u1edbi, an to\u00e0n h\u01a1n v\u00e0 hi\u1ec7u qu\u1ea3 h\u01a1n.<\/p>\n\n\n\n<p>M\u1ed9t h\u01b0\u1edbng ti\u1ebfp c\u1eadn \u0111\u1ea7y h\u1ee9a h\u1eb9n trong tr\u1ebb h\u00f3a da l\u00e0 \u1ee9ng d\u1ee5ng tr\u1ecb li\u1ec7u exosome c\u00f3 ngu\u1ed3n g\u1ed1c t\u1eeb t\u1ebf b\u00e0o g\u1ed1c m\u1ee1 (ADSCs). Exosome l\u00e0 c\u00e1c t\u00fai ngo\u1ea1i b\u00e0o k\u00edch th\u01b0\u1edbc nano, tham gia truy\u1ec1n th\u00f4ng tin gi\u1eefa c\u00e1c t\u1ebf b\u00e0o th\u00f4ng qua v\u1eadn chuy\u1ec3n protein, lipid v\u00e0 axit nucleic. Kh\u00e1c v\u1edbi li\u1ec7u ph\u00e1p t\u1ebf b\u00e0o, exosome l\u00e0 m\u1ed9t chi\u1ebfn l\u01b0\u1ee3c \u201ckh\u00f4ng s\u1eed d\u1ee5ng t\u1ebf b\u00e0o\u201d, c\u00f3 th\u1ec3 gi\u1ea3m b\u1edbt r\u00e0o c\u1ea3n s\u1eed d\u1ee5ng v\u00e0 c\u1ea3i thi\u1ec7n t\u00ednh an to\u00e0n. Exosome t\u1eeb t\u1ebf b\u00e0o g\u1ed1c m\u1ee1 (ADSC-Exo) \u0111\u1eb7c bi\u1ec7t \u0111\u01b0\u1ee3c quan t\u00e2m trong da li\u1ec5u, do \u0111\u00e3 \u0111\u01b0\u1ee3c ghi nh\u1eadn kh\u1ea3 n\u0103ng gi\u1ea3m ho\u1ea1t \u0111\u1ed9ng \u03b2-galactosidase li\u00ean quan l\u00e3o h\u00f3a (SA-\u03b2-gal), \u1ee9c ch\u1ebf bi\u1ec3u hi\u1ec7n c\u00e1c protein li\u00ean quan l\u00e3o h\u00f3a v\u00e0 th\u00fac \u0111\u1ea9y t\u1ed5ng h\u1ee3p th\u00e0nh ph\u1ea7n ch\u1ea5t n\u1ec1n ngo\u1ea1i b\u00e0o (ECM) quan tr\u1ecdng, ti\u00eau bi\u1ec3u l\u00e0 collagen. Th\u00f4ng qua c\u00e1c c\u01a1 ch\u1ebf n\u00e0y, ADSC-Exo c\u00f3 ti\u1ec1m n\u0103ng ph\u1ee5c h\u1ed3i t\u00ednh to\u00e0n v\u1eb9n c\u1ee7a da b\u1ecb suy y\u1ebfu b\u1edfi qu\u00e1 tr\u00ecnh l\u00e3o h\u00f3a v\u00e0 c\u00e1c t\u00e1c \u0111\u1ed9ng m\u00f4i tr\u01b0\u1eddng.<\/p>\n\n\n\n<p>Nghi\u00ean c\u1ee9u g\u1ea7n \u0111\u00e2y cho th\u1ea5y \u0111i\u1ec1u ki\u1ec7n thi\u1ebfu oxy (n\u1ed3ng \u0111\u1ed9 O2 d\u01b0\u1edbi 5%) c\u00f3 th\u1ec3 l\u00e0m t\u0103ng \u0111\u00e1ng k\u1ec3 \u0111\u1eb7c t\u00ednh tr\u1ecb li\u1ec7u c\u1ee7a t\u1ebf b\u00e0o g\u1ed1c. Khi \u0111\u01b0\u1ee3c nu\u00f4i c\u1ea5y trong \u0111i\u1ec1u ki\u1ec7n thi\u1ebfu oxy, t\u1ebf b\u00e0o g\u1ed1c m\u1ee1 (ADSCs) th\u1ec3 hi\u1ec7n kh\u1ea3 n\u0103ng s\u1ed1ng s\u00f3t v\u00e0 t\u0103ng sinh t\u1ed1t h\u01a1n, \u0111\u1ed3ng th\u1eddi t\u1ea1o ra exosome (hypADSC-Exo) v\u1edbi th\u00e0nh ph\u1ea7n ph\u00e2n t\u1eed \u0111\u1eb7c bi\u1ec7t kh\u00e1c bi\u1ec7t so v\u1edbi khi nu\u00f4i \u1edf \u0111i\u1ec1u ki\u1ec7n b\u00ecnh th\u01b0\u1eddng. Nh\u1eefng exosome \u0111\u01b0\u1ee3c t\u1ea1o ra trong \u0111i\u1ec1u ki\u1ec7n thi\u1ebfu oxy c\u00f3 th\u1ec3 ch\u1ee9a c\u00e1c microRNA v\u00e0 protein b\u1ea3o v\u1ec7, gi\u00fap ch\u1ed1ng l\u1ea1i stress oxy h\u00f3a, k\u00edch th\u00edch t\u0103ng sinh m\u1ea1ch m\u00e1u v\u00e0 c\u00f3 th\u1ec3 th\u00fac \u0111\u1ea9y qu\u00e1 tr\u00ecnh t\u00e1i t\u1ea1o da hi\u1ec7u qu\u1ea3 h\u01a1n so v\u1edbi exosome t\u1ea1o ra \u1edf \u0111i\u1ec1u ki\u1ec7n th\u01b0\u1eddng. C\u1ee5 th\u1ec3, nh\u00f3m t\u00e1c gi\u1ea3 xem x\u00e9t s\u1ef1 \u0111i\u1ec1u ch\u1ec9nh c\u1ee7a c\u00e1c y\u1ebfu t\u1ed1 \u0111i\u1ec1u h\u00f2a l\u00e3o h\u00f3a p16 v\u00e0 p21, \u0111\u1ed3ng th\u1eddi \u0111o m\u1ee9c \u0111\u1ed9 ho\u1ea1t \u0111\u1ed9ng c\u1ee7a SA-\u03b2-galactosidase \u0111\u1ec3 x\u00e1c \u0111\u1ecbnh kh\u1ea3 n\u0103ng exosome c\u00f3 th\u1ec3 \u201c\u0111\u1ea3o ng\u01b0\u1ee3c\u201d hi\u1ec7n t\u01b0\u1ee3ng l\u00e3o h\u00f3a \u1edf t\u1ebf b\u00e0o.<\/p>\n\n\n\n<p>\u1ede ph\u1ea7n in vivo, nh\u00f3m t\u00e1c gi\u1ea3 s\u1eed d\u1ee5ng m\u00f4 h\u00ecnh chu\u1ed9t b\u1ecb l\u00e3o h\u00f3a da do ti\u1ebfp x\u00fac UV m\u00e3n t\u00ednh, m\u00f4 ph\u1ecfng t\u00ecnh tr\u1ea1ng l\u00e3o h\u00f3a do \u00e1nh s\u00e1ng \u1edf ng\u01b0\u1eddi. Qu\u00e1 tr\u00ecnh b\u00f4i hypADSC-Exo t\u1ea1i ch\u1ed7 \u0111\u01b0\u1ee3c th\u1ef1c hi\u1ec7n 2 l\u1ea7n m\u1ed7i tu\u1ea7n trong 6 tu\u1ea7n, k\u00e8m theo \u0111\u00e1nh gi\u00e1 \u0111\u1ecbnh k\u1ef3 v\u1ec1 h\u1ec7 s\u1ed1 nh\u0103n, \u0111\u1ed9 d\u00e0y da, \u0111\u1ed9 \u1ea9m, c\u00f9ng \u0111\u1ed9 \u0111\u00e0n h\u1ed3i\u2014c\u00e1c ch\u1ec9 s\u1ed1 ch\u00ednh c\u1ee7a t\u00ecnh tr\u1ea1ng da. K\u1ef9 thu\u1eadt nhu\u1ed9m m\u00f4 h\u1ecdc nh\u01b0 Hematoxylin &amp; Eosin (H&amp;E) v\u00e0 Masson\u2019s Trichrome \u0111\u01b0\u1ee3c d\u00f9ng \u0111\u1ec3 \u0111o \u0111\u1ecbnh l\u01b0\u1ee3ng \u0111\u1ed9 d\u00e0y c\u1ee7a l\u1edbp bi\u1ec3u b\u00ec, l\u1edbp b\u00ec v\u00e0 m\u1eadt \u0111\u1ed9 collagen. B\u00ean c\u1ea1nh \u0111\u00f3, nh\u00f3m t\u00e1c gi\u1ea3 c\u0169ng ph\u00e2n t\u00edch bi\u1ec3u hi\u1ec7n gene c\u1ee7a collagen lo\u1ea1i I v\u00e0 III, c\u0169ng nh\u01b0 c\u00e1c metalloproteinase (MMP) 1, 2 v\u00e0 3, \u0111\u1ec3 l\u00e0m r\u00f5 c\u00e1ch hypADSC-Exo \u1ea3nh h\u01b0\u1edfng \u0111\u1ebfn c\u00e1c con \u0111\u01b0\u1eddng ph\u00e2n t\u1eed d\u1eabn \u0111\u1ebfn l\u00e3o h\u00f3a da.<\/p>\n\n\n\n<p>K\u1ebft qu\u1ea3 c\u1ee7a nghi\u00ean c\u1ee9u cho th\u1ea5y hypADSC-Exo c\u00f3 th\u1ec3 \u0111i\u1ec1u ch\u1ec9nh t\u00edch c\u1ef1c l\u00e3o h\u00f3a da do \u00e1nh s\u00e1ng b\u1eb1ng nhi\u1ec1u c\u01a1 ch\u1ebf.
